The Biblioteca Histórica de Santa Cruz houses the old bibliographic collection of the University of Valladolid. It holds collections from different provenances due to numerous donations and bequests that have increased its original collection over the centuries. The bibliographic fonds is organised into four sections: 1) Manuscripts, 2) Incunabula and Rare Books, 3) Printed matter, and 4) Miscellaneous files.
The Incunabula and Rare Books collection is composed of 528 works bound in 350 volumes, of which 203 are incunabula. This collection includes a Hebrew incunabulum printed in Leiria in 1495 by Abraham ben Samuel d'Ortas, of Jacob Ben Asher's Tur Orah Hayyim. This volume entitled "Semina vitae, seu primus ordo" (U/Bc IyR 334) contains a description of ceremonies conducted at the synagogue, among other themes. A digital copy is available online.

The origins of the Colegio de Santa Cruz Library date back to the foundation of this College by Cardinal Mendoza in 1483. The library was placed in a large room on the first floor of the College. An inventory drawn up in 1589-90 reveals that the library contained 970 volumes arranged on the shelves according to subjects: Canons, Law, Arts, Medicine, and Theology. At the beginning of the 18th century, the old library was remodelled and acquired the appearance it has today, with its furniture made by Alonso del Manzano in 1705. The library was organised horizontally in two bodies joined by a staircase and vertically in sections separated by Solomonic columns.
