The Handschriften collection of the University and State Library of Saxony-Anhalt comprises an extensive collection of manuscripts, including about 400 medieval occidental manuscripts, over 550 musical manuscripts, oriental and modern manuscripts of various origins, 1,600 incunabula, 350 rare books, and approximately 4,000 documents.
It is part of this collection the so-called "Halle manuscript", a 381-folio manuscript discovered by Henry Charles Lea in 1902 that documents 1,046 cases tried by the Inquisition in Toledo from 1575 to 1610 (MSS Yc 2º 20 (1)). It comprises trial summaries sent to the Suprema, the High Court of the Spanish Inquisition in Madrid. The volume is organised chronologically and was published by Julio Serra (2005). Another Sephardic-related item of this collection is a partial copy of the Latin version of Moses Raphael de Aguilar's "Reposta ahum Papel, qui aqui mandou de França uma pesoa de nossa naçao" (Answer to a paper that was sent here by a person of our nation) translated by Johann Christoph Wolf (1683-1739) (Misc.Quart. 20 a).

The Halle University Library was founded in 1696. In 1817-18, parts of the Wittenberg University Library (founded in 1502) were transferred to the Halle University Library.
Otto Hartwig (1830-1903) became the first full-time director of the university library in 1876, managing an important renovation in terms of redesigning catalogues and the library system. In 1878-80, a new library building was built in August-Bebel-Straße 50.
The university library assumed the functions of a state library in 1948. In 1954, the building of the former Oberbergamt (Mining Office), in August-Bebel-Straße 13, was acquired.
In 1969-1971, a new uniform library system was created. New innovations came later when, in 1991, the electronic cataloguing system was started.
In 1996, the library buildings underwent a large rehabilitation, which lasted until 1999.
